An Experimental Study on Liver Regeneration and Assessment of Liver Regeneration Using Technitium 99m DISIDA Following Massive Hepatectomy


Abstract
The liver has the capacity to undergo compensatory growth and to regulate it size. Most of the studies of regeneration have been done in the regenerating liver after partial hepatectomy, because partial hepatectomy is being performed with some
frequency
for neoplasm and trauma of liver. This study was performed to demonstrate some data concerning regeneration ratio, regenerative features and assessment of liver function using 99m-technetium DISIDA plasma clearance rate (DISIDA-K) after massive
hepatectomy.
Healthy adult NewZealand While rabbits weighing about 2 kg were divided into two groups ; Sham operation group as control(n=10) and 75% hepatectomy group(MH group, n=35). Healthy 5 rabbits among all control rabbits were performed H-E stain, PAS
stain,
MGP stain and hepatobiliary scintigram using 99mTc-DISIDA for calculating DISIDA-K.
All twenty rabbits of control group were sacrified and removed total liver. The removed liver of each control rabbit was divided as right lobe, central lobe, caudate lobe and left lobe, weighed and calculated percentage of each lobe out of total
liver.
In MH group, 5 rabbits were sacrificed at immediate time after operation (o time), 1 day, 3 days, 7 days, 14 days, 21 days and 28 days, and performed hepatobiliary scintigrams using 99mTc-DISIDA for calculating DISIDA-KJ. Author calculated
regeneration
ration and performed H-E stains, PAS stains and MGP stains in MH group except 0-time rabbits.
@ES The results obtained were summarized as follows:
@EN 1) Each liver of control group was composed as left lobe, central lobe, right lobe and caudate lobe, and the mean percentages of each lobes were 25.1%, 52.1%, 17.0% and 5.7% respectively.
2) The mean regeneration ratios of residual livers were 33.8% in 1 days, 49.5% in 3 days, 93.1% in 7 days, 100.9% in 14 days, 114.9% in 21 days and 114.4% in 28 days after MH.
3) Mitotic figures, dark cells and double nucleated cells in MH group reached peak level at 3 days and returned to normal level at 14-21 days after MH. Positive PAS reactios decreased significantly form 1 day to 3 day and were comparable to
control
level at 7 days after MH. The MGP reacting granules were beginning to accumulate on the parenchymal cells around the portal space, attaining a peak at 3 days and returned to normal level at 21 days after MH.
4) In control group, the mean DISIDA-K(%) was 1.51 min-1(100%), and in MH group, the mean DISIDA-K was 0.46 min-1(30.5%) at 0time, 0.74 min-1(49.0%) at 1 day, 1.03 min-1 (68.2%) at 3 days, 1.08 min-1(71.5%)at 7 days, 1.17 min-1 (77.5%) at 14
days,

min-1(84.1%) at 21 days and 1.30 min-1(86.1%) at 28 days.
